The microfold (M) cells of the Peyer’s patches (PP) are specialized antigen-sampling cells of the gut-associated lymphoid tissue (GALT) and M cells have been identified to mediate an essential step in oral vaccine delivery. As M cells play a central role in the uptake of antigen and in the initiation of T-cell dependent immune responses, we determined the role of MHC-II molecules in the inductive phase of mucosal immunity. Using a murine model that lacks MHC-II molecules on T cells due to a targeted disruption of the invariant chain gene (I-Aalpha) (I-Aalpha(mut)) mice we analyzed the cellular composition of the mucosal immune system of the small and large intestine and compared this with the cellular composition in normal mice (WT) and in mice deficient for MHC-II molecules only on B cells (beta(mut)) to address the question whether T cells are involved in the uptake of antigen by M cells. Cellular composition in the small and large intestine was analyzed by flow cytometry and laser scanning cytometry. The function of isolated Peyer’s patch M cells as antigen-presenting cells (APC) was studied in cell culture experiments. We found that the numbers of M cells were reduced in beta(mut) mice compared to WT mice and that M cells were also reduced in I-Aalpha(mut) mice. In I-Aalpha(mut) mice, there was no difference in cell composition in the small intestine. In the large intestine, a decrease in T-helper cells was observed in the I-Aalpha(mut) mice. These data demonstrate that M cells of the small intestine rely on a functional MHC-II expression on T cells.1. Field of the Invention
